SS-31 (Elamipretide) — Mitochondrial Cardiolipin-Targeted Tetrapeptide
SS-31, also known as elamipretide or MTP-131, is a synthetic aromatic-cationic tetrapeptide engineered to selectively localize to the inner mitochondrial membrane through electrostatic interaction with cardiolipin. Developed by Hazel Szeto at Weill Cornell Medicine, SS-31 has been characterized in more than fifteen years of preclinical and clinical research on mitochondrial dysfunction in cardiac, renal, and neurological models.
Mechanism of Action
SS-31 binds cardiolipin at the inner mitochondrial membrane, stabilizing cristae architecture and preserving electron-transport-chain efficiency under oxidative stress. Preclinical studies show it reduces reactive oxygen species production, improves ATP synthesis under ischemic conditions, and preserves mitochondrial respiratory capacity in aged tissues. Its mechanism is distinct from generic antioxidants: rather than scavenging free radicals stoichiometrically, SS-31 modifies the redox environment of the membrane itself, which has been the central focus of research on its activity in heart-failure, ischemia-reperfusion, and Barth-syndrome models.
